The glycinate form is chelated to the amino acid glycine, which independently lowers core body temperature (a required trigger for sleep onset) and stabilizes non-REM sleep. Glycinate also absorbs at roughly 40 to 80 percent compared to the 4 percent you get from cheap magnesium oxide, which matters when you are taking it nightly.
Valerian Root contributes valerenic acid, a positive allosteric modulator of GABA-A receptors. It binds at a different site than benzodiazepines, which is why it calms without the blunting, the amnestic effect, or the withdrawal profile. Clinical evidence is modest but consistent: people fall asleep faster and report better subjective sleep quality with nightly use over two to four weeks. It is not a sedative, it is a downshift.
Ashwagandha Root addresses the upstream problem most people never name. Sleep disruption in adults under chronic stress is rarely about sleep mechanics, it is about cortisol. A 2021 meta-analysis of five randomized controlled trials (Cheah et al., PLOS One) found ashwagandha extract produced a standardized mean difference of -0.59 on overall sleep, with specific improvements in sleep onset latency, total sleep time, and mental alertness on rising. The mechanism is HPA-axis modulation, which is a clinical way of saying the nervous system stops bracing for threats that are not coming.
Reishi (Ganoderma lucidum) is the oldest entry in the stack, used for more than two thousand years in East Asian medicine specifically for what practitioners called "settling the spirit."
